Don's drug replenishes the endogenous deficiency of glucosamine , enhances the synthesis of proteoglycans , synovial fluid and hyaluronic acid .
It has an analgesic and anti-inflammatory effect, normalizes the permeability of the joint capsule and enzymatic processes in the cellular structures of articular cartilage and synovial membrane.
In the process of producing chondroitinsulfuric acid, the drug improves the process of sulfur and normalizes calcium , slows down degenerative processes in joints, and reduces joint pain.
Pharmacokinetics
After oral administration, the drug is completely and quickly absorbed in the small intestine (absorption - 90%). Easily overcomes biological barriers, distributed mainly in the cartilage tissue of joints. The half-life is 68 hours. It is excreted unchanged in urine and partially in feces.
Pharmacological properties of the drug Dona
Pharmacodynamics . A drug that affects metabolic processes in cartilage tissue. Glucosamine sulfate is a salt of the natural aminomonosaccharide glucosamine with low molecular weight, carefully purified from macromolecular components. The drug replenishes the endogenous deficiency of glucosamine, stimulates the synthesis of proteoglycans and hyaluronic acid in synovial fluid. Increases the permeability of the joint capsule, restores enzymatic processes in the cells of the synovial membrane and articular cartilage. The anti-inflammatory effect of glucosamine sulfate is due to blocking the formation of superoxide radicals by macrophages and inhibition of lysosomal enzymes, interleukin 1, collagenase, phospholipase A2, which leads to a slowdown in structural damage to articular cartilage, which has been repeatedly proven in long-term clinical studies. The drug promotes the fixation of sulfur in the process of chondroitinsulfuric acid synthesis and normal calcium deposition in bone tissue. Inhibits the development of degenerative processes, reduces the severity of joint pain and restores their function. Sulfates are also involved in the synthesis of glycosaminoglycans and the metabolism of cartilage tissue. The sulfate ester side chains of proteoglycans play an important role in maintaining the elasticity and water retention capacity of the cartilage matrix. A decrease in the severity of clinical symptoms is usually noted 2 weeks after the start of treatment, with clinical improvement continuing for 8 weeks after discontinuation of the drug. The results of clinical studies of continuous therapy with the drug for 3 years indicate a progressive increase in its effectiveness, expressed in regression of disease symptoms and slowdown of structural damage to articular cartilage, confirmed by X-ray. Glucosamine sulfate is well tolerated, with no significant effects on the cardiovascular, respiratory or central nervous system. Contraindications
The drug in powder form, due to the content of aspartame , is contraindicated for phenylketonuria .

Dona, instructions for use (Method and dosage)
Take Don's drug for joints in powder form orally 30 minutes before meals, after dissolving 1 packet in a glass of water once a day for 1-3 months. Repeated course - after 2 months.
Don tablets are taken simultaneously with meals, 1 or 2 tablets three times a day.
Don's solution for intramuscular administration: an ampoule of the drug in one syringe is mixed with an ampoule of solvent. Inject deeply intramuscularly three times a week for 1-1.5 months.
To increase the effectiveness of treatment, it is advisable to combine parenteral administration of Dona with oral administration of the drug.

Pharmacological properties

Pharmacokinetics
When administered intramuscularly, bioavailability is 95%, rapidly distributed in tissues, half-life of about 60 hours, excreted mainly by the kidneys.
Indications for use
Primary and secondary osteoarthritis, osteochondrosis, spondyloarthrosis.
Contraindications
Individual hypersensitivity to glucosamine, lidocaine hydrochloride and other components of the drug. Due to the presence of lidocaine in the product, it is contraindicated in patients with cardiac conduction disorders and acute heart failure, a history of epileptiform seizures, severe liver and kidney dysfunction; during pregnancy and lactation, as well as in children under 12 years of age.
Carefully
The drug should be used with caution in patients with chronic heart failure and arterial hypotension. Caution should be exercised in patients with allergies to seafood (shrimp, shellfish).
Directions for use and doses
Intramuscularly! The drug is not intended for intravenous administration.
Before use, mix solution B (solvent) with solution A (drug solution) in one syringe. The prepared solution of the drug is administered intramuscularly in a dose of 3 ml (solutions A + B) 3 times a week for 4-6 weeks. Injections of the drug can be combined with oral administration of the drug in powder to prepare an oral solution (sachets). The duration and regimen of treatment is prescribed by the attending physician.
Side effect
The drug is well tolerated. In some cases, possible: flatulence, diarrhea, constipation, allergic reactions - urticaria, itching. Due to the content of lidocaine in the drug, the following are possible: nausea, vomiting, drowsiness, diplopia, headache, dizziness, numbness of the tongue and oral mucosa, tremor, euphoria, disorientation, cardiac conduction disturbances.
Overdose

Interaction with other drugs
Compatible with non-steroidal anti-inflammatory drugs, paracetamol and glucocorticosteroids.
Caution should be exercised when prescribing injections of the drug in combination with beta-blockers, digitoxin, ajmaline, amiodarone, verapamil, quinidine, procainamide, hexenal or sodium thiopental, MAO inhibitors, polymyxin-B, hypnotics or sedatives, cimetidine.
